miR-20a attenuates acute lung injury in septic rats via targeting TLR4

Background: Sepsis is most likely to cause lung damage in patients, and the detection rate mortality are high. Here, we investigated expression of miR-20a sepsis-induced acute injury (ALI) rats its effect on inflammatory response, reveal possible molecular mechanism. Method: The model caused by sepsis was established cecal ligation puncture. tissue determined RT-qPCR. Acute were injected with 5 nmol agomir or NC every day for 3 days. Rats sacrificed arterial bleeding tissues removed. Serum interleukin (IL) -1β, IL-6, tumor necrosis factor alpha (TNF-α) detected ELISA. HE staining used observe pathology calculate pathological score injury. Western blot determine level TLR4 nuclear transcription κB p65 (NF-κB p65) protein tissue. luciferase reporter assay verify binding non-coding TLR4. Results: We found that compared Normal group, ALI decreased (p < 0.05). In plasma IL-1β, TNF-α significantly lower than group 0.05), while higher those results showed Compared NF-κB 0.01). reporting experiment confirmed a target gene miR-20a. Conclusion: To sum up, exerts protective through anti-inflammatory effect. targeting may be an effective method reduce response ALI.
